      背景技術(shù)
      :口蹄疫是由口蹄疫病毒(FoofflwJ-moW/zWn,FMDV)弓I起偶蹄動物的一種急性、高度接觸性、發(fā)熱性傳染病,以傳播迅速、感染率高而著稱。該病一旦爆發(fā)將對發(fā)病國造成巨大的經(jīng)濟(jì)損失,世界各國對該病的研究極為重視,國際獸醫(yī)局將其列為A類傳染病之首??谔阋邔儆谛NA病毒科口蹄疫病毒屬的成員,有A、O、C、Asial、SAT1、SAT2禾BSAT3型7個血清型。目前我國口蹄疫的防制仍以預(yù)防為主。盡管傳統(tǒng)疫苗在口蹄疫預(yù)防控制中仍占主導(dǎo)地位,但生產(chǎn)成本昂貴,免疫期短,疫苗制備過程中病毒逃逸等危險因素很難避免。因此,傳統(tǒng)疫苗亟需加以改進(jìn),研制安全、高效的口蹄疫分子疫苗仍顯得非常必要。隨著生物技術(shù)、基因工程等高新技術(shù)的發(fā)展,人們意識到通過基因工程的手段,利用生物反應(yīng)器來高效表達(dá)口蹄疫基因,可望達(dá)到大幅度提高口蹄疫抗原產(chǎn)量、降低生產(chǎn)成本的目的(ConneelyO.M.,BiotechnologyintheFeedIndustry,T.P.Lyons(Ed),AlltechTechnicalPublications.Nicholasville,KY.57-66,1992)。桿狀病毒表達(dá)系統(tǒng)這一生物反應(yīng)器是八十年代建立起來的。自從1983年首次利用桿狀病毒表達(dá)系統(tǒng)高效表達(dá)了人的a-干擾素以來(Smith,Mol.CellBiol.,3:2156-2165,1983),已有數(shù)十個外源基因得到了高效表達(dá),僅我國就有a-干擾素(楊冠珍等,生物化學(xué)與生物物理學(xué)報,22:355-361,1990)、慈菇蛋白酶抑制劑(季平等,蠶業(yè)科學(xué),21:223-227,1995)、馬立克氏病毒糖蛋白B(肖慶利等,蠶業(yè)科學(xué),23:104-108,1997)等多種。利用此系統(tǒng)來生產(chǎn)口蹄疫抗原的優(yōu)點在于1.這一表達(dá)系統(tǒng)的表達(dá)效率極高,產(chǎn)量可達(dá)10毫克級/蟲的水平。因而可大大降低口蹄疫抗原的生產(chǎn)成本并使通過基因工程方法大規(guī)模生產(chǎn)口蹄疫抗原成為可能;2.這一表達(dá)系統(tǒng)為真核表達(dá)系統(tǒng),其表達(dá)的外源蛋白質(zhì)可進(jìn)行翻譯后修飾,使其在生化性質(zhì)和生物活性等與天然產(chǎn)品相似,這為所表達(dá)的口蹄疫抗原具有正常的蛋白結(jié)構(gòu)和生物學(xué)免疫活性提供了保證。目前,桿狀病毒表達(dá)系統(tǒng),尤其是其中的家蠶(Bm)-家蠶桿狀病毒(BmNPV)表達(dá)系統(tǒng)是世界上最具有商業(yè)開發(fā)價值的真核生物個體表達(dá)系統(tǒng)之一。
      發(fā)明內(nèi)容本發(fā)明所要解決的技術(shù)問題是克服現(xiàn)有技術(shù)的不足,提供一種利用桿狀病毒表達(dá)系統(tǒng)在昆蟲體內(nèi)安全、高效的表達(dá)口蹄疫抗原的方法。本發(fā)明所要解決的技術(shù)問題是通過以下技術(shù)方案來實現(xiàn)的一種制備口蹄疫抗原的方法,包括將口蹄疫不同的基因組合分別克隆到桿狀病毒運載載體中,獲得轉(zhuǎn)移載體;用所獲得的轉(zhuǎn)移載體轉(zhuǎn)染桿狀病毒進(jìn)行DNA重組,獲得重組桿狀病毒;用重組桿狀病毒感染昆蟲宿主;培養(yǎng)被感染的昆蟲宿主使其進(jìn)行口蹄疫抗原表達(dá);收獲并純化所表達(dá)的抗原。其中,所述的口蹄疫不同的基因組合優(yōu)選自SEQIDNO:l、SEQIDNO:3或SEQIDNO:5所示的堿基序列;所述的桿狀病毒運載載體優(yōu)選自AcRP23-/acZ,AcRP6-SC,AcUWl-/acZ,BacPAK6,BactoPac,Bacmid,BlucBacII(pETL),p2Bac,p2Blue,p89B310,pAc360、pAc373,pAcAB3、pAcAB4,PAcAS3,pAcC129,pAcC4,DZI,pAcGP67,pAcIEl,pAcJPl,pAcMLF2、pAcMLF7、pAcMLF8,pAcMPl、pAcMP2,pAcRP23、pAcRP25,pAcRW4,pAcsMAG,pAcUWl、pAcUW21、pAcUW2A、pAcUW2B、pAcUW3、pAcUW31、pAcUW41、pAcUW42、pAcUW43、pAcUW51,pAcVC2、pAcVC3,pAcYMl,pAcJcC5,pBacl、pBac2,pBlueBacIII,pBlueBacHis,pEV55、mXIV,pIEINeo,pJVETL,pJVNhel,pJVP10,pJVrsMAG,pMBac,pP10,pPAKl,pPBac,pSHONEX1.1,pSYNXIVVI+,pSYNVI+wp,pSYNXIVVI-,pVL1391、pVL1392、pVL1393、pVL941、pVL945、pVL985,pVTBac,pBM030,pUAC-5或其它類似的桿狀病毒同源重組或轉(zhuǎn)座載體,更優(yōu)選為pVL1393。所構(gòu)建的轉(zhuǎn)移載體優(yōu)選為pVL1393(P1-2A3C)、pVL1393(ORF)或pVL1393(VPl)。所述的桿狀病毒選自BmNPV、AcMNPV、ApNPV、BsSNPV、CfMNPV、EoSNPV、HaNPV、HzNPV、LdMNPV、MbMNPV、OpMNPV、S1MNPV、SeMNPV、或TeNPV,優(yōu)選為家蠶桿狀病毒Bm-NPV-ZJ8;所述的重組桿狀病毒優(yōu)選為以下任意一種(1)家蠶核型多角體病毒rBmNPV(ORF),其微生物保藏號是CGMCCNO.1980;保藏時間是2007年3月20H;保藏單位是中國微生物菌種保藏管理委員會普通微生物中心;保藏地址是北京市海淀區(qū)中關(guān)村北一條13號,中國科學(xué)院微生物研究所;(2)家蠶核型多角體病毒rBmNPV(P1-2A3C),其微生物保藏號是CGMCCN0.1979;保藏單位是中國微生物菌種保藏管理委員會普通微生物中心;保藏地址是北京市海淀區(qū)中關(guān)村北一條13號,中國科學(xué)院微生物研究所;(3)家蠶核型多角體病毒rBmNPV(VPl),其微生物保藏號是CGMCCN0.1975;保藏單位是中國微生物菌種保藏管理委員會普通微生物中心;保藏地址是北京市海淀區(qū)中關(guān)村北一條13號,中國科學(xué)院微生物研究所。所述的昆蟲宿主選自包括家蠶(Bombyxmori)、野蠶(Bombyxmandarina)、蓖麻蠶(Philosamiacynthiaricim)、樟蠶(Dictyoplocajapanica)、樗蠶(Philosamiacynthiapryeri)、柞蠶(Antheraeapernyi)、日本柞蠶(Antheraeayamamai)、野天蠶(Antheraeapolyphymus)、苜蓿尺蠖(Atographacaliforica)、茶尺蠖(Ectropisobliqua)、甘蘭夜蛾(Mamestrabrassicae)、斜紋夜蛾(Spodopteralittoralis)、秋粘蟲(Spodopterafrugiperda)、粉紋夜蛾(Trichoplusiani)、行軍蟲(Thaumetopoeawilkinsoni)、棉鈴蟲(Heliothisarmigera)、美國棉鈴蟲(Heliothiszea)、煙青蟲(Heliothisassulta)、'煙草夜蟲我(Heliothisvirescens)、東方茅占蟲(Pseudaletiaseparata)、舞毒蛾(Lymantriadispar)等;更優(yōu)選為家蠶(Bombyxmori)。所述的感染是指重組桿狀病毒通過口食或透過表皮來感染1-5齡的昆蟲幼蟲或蛹體(更優(yōu)選為將重組家蠶桿狀病毒感染家蠶細(xì)胞或穿刺接種l-5齡的家蠶幼蟲或蛹,在感染3-6天后收集含口蹄疫抗原的家蠶幼蟲或蛹的體液或組織勻漿);其中,所述的蛹體為l-2天的早期嫩蛹。本發(fā)明采用基因重組技術(shù),將來源于口蹄疫病毒的不同基因組合,包括P1-2A3C、ORF、VP1構(gòu)建到各種桿狀病毒運載載體(如AcRP23-lacZ,AcRP6-SC,AcUWl-lacZ,BacPAK6,BactoPac,Bacmid,BlueBacII(pETL),p2Bac,p2Blue,p89B310,pAc360、373,pAcAB3、4,pAcAS3,pAcC129、C4、DZ1,pAcGP67,pAcIEl,pAcJPl,pAcMLF2、7、8,pAcMPl、2,pAcRP23、25,pAc證4,pAcsMAG,pAcUWl、21、2A、2B、3、31、41、42、43、51,pAcVC2、3,pAcYMl,pAcJcC5,pBacl、2,pBlueBacIII,pBlueBacHis,pEV55、mXIV,pIEINeo,pJVETL,pJVNhel,pJVP10,pJVrsMAG,pMBac,pP10,pPAKl,pPBac,pSHONEX1.1,pSYNXIVVI+,pSYNVI+wp,pSYNXIVVI-,pVL1391、1392、1393,pVL941、945、985,pVTBac,pBM030,pUAC-5)上,使口蹄疫病毒不同的基因組合,包括ORF、P1-2A3C、VP1基因在多角體啟動子、p10啟動子或別的病毒和真核生物的強啟動子控制之下,通過體內(nèi)或體外(invivo/invitro)重組,將口蹄疫病毒不同的基因組合,包括P1-2A3C、ORF、VP1整合到桿狀病毒的基因組上,得到重組病毒。重組病毒可通過經(jīng)口食下或采用各種手段透過表皮感染l-5齡(最優(yōu)時間為四或五齡)的昆蟲幼蟲或蛹體(最優(yōu)時間為l-2天的早期嫩蛹),表達(dá)生產(chǎn)口蹄疫病毒抗原。本發(fā)明中最優(yōu)選的一個技術(shù)方案是將SEQIDNO:l(P1-2A3C)、SEQIDNO:3(ORF)或SEQIDNO:5(VP1)所示的堿基序列插入到運載載體pVL1393上,再通過體內(nèi)重組將全長P1-2A3C、ORF、VP1基因轉(zhuǎn)移到家蠶桿狀病毒親本株Bm-NPV-ZJ8的基因組上,替代基因組上的Polyhedrin基因,通過空斑篩選技術(shù)和PCR檢測技術(shù),獲得攜帶口蹄疫不同基因組合的重組家蠶桿狀病毒rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)。將其感染家蠶細(xì)胞系或穿刺接種1-5齡的家蠶幼蟲或蛹,大量繁殖rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)。當(dāng)rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)在蠶體內(nèi)復(fù)制時,ORF、P1-2A3C、VP1在多角體蛋白基因(polh)啟動子控制下表達(dá),產(chǎn)生口蹄疫抗原。在感染3-6天(最佳為5天)后收集含口蹄疫抗原的家蠶幼蟲或蛹的體液(或整體勻漿),每毫升蠶血淋巴可產(chǎn)生IO毫克以上的口蹄疫抗原,殺滅感染性病原后,經(jīng)過蛋白純化后便得到安全、高效的口蹄疫抗原,此種抗原可用于制備疫苗。本發(fā)明方法采用桿狀病毒表達(dá)系統(tǒng)在家蠶生物反應(yīng)器中安全、高效的生產(chǎn)口蹄疫抗原,其生產(chǎn)成本顯著低于傳統(tǒng)的制備口蹄疫抗原方法(例如通過細(xì)胞繁殖病毒制備口蹄疫抗原),無需投資建廠,無三廢,電力和水資源等能源消耗極少。由于家蠶已經(jīng)我國衛(wèi)生部批準(zhǔn)為食藥兼用昆蟲,所以將本發(fā)明方法所制備的抗原純化后,安全性極高,可直接制作疫苗免疫動物。總體而言,本發(fā)明方法可以大幅度降低口蹄疫抗原的生產(chǎn)成本,具有安全、高效、能耗少、成本低等諸多優(yōu)點。圖l、口蹄疫病毒P1-2A3C在家蠶幼蟲中的表達(dá)。圖2、口蹄疫病毒P1-2A3C在家蠶蛹中的表達(dá)。圖3、口蹄疫病毒全長ORF在家蠶幼蟲中的表達(dá)。圖4、口蹄疫病毒全長ORF在家蠶蛹中的表達(dá)。圖5、口蹄疫病毒VP1在家蠶幼蟲中的表達(dá)。圖6、口蹄疫病毒VP1在家蠶蛹中的表達(dá)。具體實施方式為了進(jìn)一步闡述本發(fā)明,下面給出一系列實施例。這些實施例完全是例證性的,它們僅用來對本發(fā)明進(jìn)行具體描述,不應(yīng)當(dāng)理解為對本發(fā)明的限制。試驗材料1.大腸桿菌株E.coliTG1和DHsa購自Promega公司;運載載體pVL1393(購自Invitrogen公司)、家蠶細(xì)胞BmN、家蠶核型多角體病毒親本株Bm-NPV-ZJ8由中國農(nóng)業(yè)科學(xué)院生物技術(shù)研究所保存;口蹄疫病毒由中國農(nóng)業(yè)科學(xué)院蘭州獸醫(yī)研究所口蹄疫國家參考實驗室保存;抗原檢測試劑盒由中國農(nóng)業(yè)科學(xué)院蘭州獸醫(yī)研究所制備,家蠶品種JY1由中國農(nóng)業(yè)科學(xué)院生物技術(shù)研究所保存。2.酶與試劑限制性內(nèi)切酶、連接酶為Promega公司產(chǎn)品。3.生化試劑IPTG、X-Gal為Promega公司產(chǎn)品。Lipofectin、低融點瓊脂糖LMP、PCR試劑盒、T4DNA連接酶、RNA酶、ProteinaseK、胎牛血清及其他試劑購于Invitrogen公司,細(xì)胞培養(yǎng)基TC-100購于Sigma公司。4.培養(yǎng)基大腸桿菌培養(yǎng)基為LB(l。/。蛋白胨、0.5%酵母提取物、l%NaCl,pH7.0);家蠶細(xì)胞培養(yǎng)基為TC-IOO。5.口蹄疫病毒基因不同組合表達(dá)產(chǎn)物的動物實驗在中國農(nóng)業(yè)科學(xué)院蘭州獸醫(yī)研究所動物隔離實驗室進(jìn)行。實施例1口蹄疫抗原的制備、純化及動物免疫實驗及病毒攻擊保護(hù)實驗1、P1-2A、3C基因的克隆和序列分析1.1目的基因的獲得設(shè)計引物,通過RT-PCR的方法擴(kuò)增出口蹄疫病毒抗原蛋白P1-2A基因與非結(jié)構(gòu)蛋白3C基因。所設(shè)計的抗原蛋白P1-2A基因和非結(jié
