背景技術:
周圍神經損傷是常見的臨床病癥,而周圍神經損傷后能夠再生。miRNACmicroRNA, 微RNA)是一類具有19 25nt的RNA分子,由其前體分子在酶的作用下加工生成,其前體序列一般為70 120nt,廣泛存在于生物體中。由于miRNA在各種生物體中具有保守性, 因此普遍認為miRNA的功能是參與生命的一些基本過程;在動物機體,miRNA通過轉錄后調控許多基因和蛋白的表達,在發(fā)育、分化、增殖、細胞的存活與癌變以及神經系統(tǒng)等發(fā)揮重要作用?;A和臨床研究表明,miRNA無論在正常生理過程還是在疾病過程中都是重要的調節(jié)因子。許多miRNA呈現(xiàn)組織特異性表達,因此在調控組織特異性和組織特異性基因的功能中發(fā)揮重大作用。近年來的研究發(fā)現(xiàn)miRNA與神經再生有著極為密切的關系,例如, miRNA在軸突生長及突觸功能和可塑性等方面具有調控作用。周圍神經損傷后,施萬細胞的增殖對周圍神經修復具有重要作用,因此作為轉錄后調控的與增殖有關miRNA的發(fā)現(xiàn)和功能鑒定對周圍神經損傷的修復具有十分重要的意義。
發(fā)明內容
由于各種意外造成的神經損傷的發(fā)病率逐年增加;而神經損傷后,再生能力差,致殘率高,因此針對神經損傷和治療的生物分子的發(fā)現(xiàn)具有重要的應用價值。本發(fā)明以大鼠坐骨神經離斷后的L4-6背根神經節(jié)和近端神經為材料進行miRNA 的Solexa測序。測序結果經過SOAP軟件與大鼠基因組序列比對,MIREAP軟件分析miRNA 前體的發(fā)夾結構,濾掉自由能大于-20 kcal/mol的候選miRNA,基于miRBaSel7.0過濾掉已報道的miRNA,MiPred軟件進一步過濾掉功能上非保守的虛擬的miRNA前體等生物信息學分析,并進一步通過RT-PCR驗證。然后對這些新的miRNA進行功能篩選,結果發(fā)現(xiàn)了增殖有關的七十個新的miRNA。本發(fā)明所提供的miRNA克隆自大鼠坐骨神經離斷后的L4-6背根神經節(jié)和近端神經,成熟miRNA序列長度約為22nt,其前體序列具有miRNA前體典型的二級結構,符合miRNA的結構特征。本發(fā)明的目的是提供七十個新的miRNA、miRNA前體和其反義寡核苷酸序列,以及該發(fā)明所涉及的miRNA、miRNA前體和其反義寡核苷酸在制備神經損傷藥物中的用途。本發(fā)明的具體技術方案如下

具體實施例方式1.動物模型構建和樣品制備
成年雄性 SD大鼠(180 220 g)被麻醉后(10 mg/kg xylazine, 95 mg/kg keUamine, acepromazine 0.7 mg/kg),左后肢外側坐骨神經被切開一個1 cm長的切口,然后皮膚被縫合。動物飼養(yǎng)在合適的溫度和濕度條件下進行,每天12 h光照,自由飲水和采食。坐骨神經切斷后的0,1,4,7和14天,處死大鼠,取大鼠坐骨神經離斷后的L4-6背根神經節(jié)和坐骨神經缺口處向上0. 5 cm的近端神經用于分析。提取和Solexa測序
用1Trizol提取總RNA,Solexa測序送北京基因組研究所(BGI)進行。18 30 nt的小RNA經過凝膠純化,連接到Solexa測序接頭,連接產物被純化、逆轉錄和PCR擴增。擴增產物經過凝膠純化后進行測序,測序得到的圖像文件被轉換成數字信號,在移去雜質信號和接頭序列后,結果被計算機分析處理。新miRNA的預測
從Solexa測序得到的小分子RNA序列首先用SOAP軟件與大鼠基因組序列比對; MIREAP用來分析候選miRNA的發(fā)夾結構;濾掉自由能大于_20 kcal/mol的候選miRNA ; 基于miRBaSel7. 0,過濾掉已報道的miRNA ;MiPred進一步過濾掉功能上非保守的虛擬的 miRNA 前體(pre-miRNA)。RT-PCR 分析
同Solexa測序的小分子RNA用來進行定量RT - PCR分析;10 ng的RNA用莖環(huán)逆轉錄引物和TaqMan反轉錄試劑盒(ABI)逆轉錄到cDNA。然后進行PCR實驗,用三蒸水作為陰性模板對照,進行PCR分析。對施萬細胞增殖的影響
根據miRNA成熟體的序列,分別設計合成了它們的mimic及inhibitor,用DMEM培養(yǎng)基培養(yǎng)施萬細胞,利用Lipofectamine RNAiMAX (invitrogen)反轉入細胞,M孔板 mimic用量為6 pmol, inhibitor用量為12 pmol,培養(yǎng)液為無血清的DMEM,按說明書操作。5-8小時后加含10%血清的完全培養(yǎng)液。培養(yǎng)36小時后,進行增殖實驗如下運用BrdU ELISA KIT (Roche)進行增殖實驗。具體實驗步驟根據說明書,簡單描述如下,將轉染36 小時的96孔板中的細胞中加入BrdU標記液,孵育4 h后固定,經一抗及二抗孵育后Leica DC 300F正置熒光顯微鏡進行觀察和拍照,隨機選取10個視野計數細胞個數;也可以酶標儀上檢測其OD值,間接反映增殖細胞數。
