本發(fā)明屬于植物除草劑抗性領(lǐng)域,具體而言,本發(fā)明涉及能賦予植物(尤其是小麥)除草劑抗性(耐受性)的蛋白及其在植物育種和雜草管理中的應(yīng)用。
背景技術(shù):
在農(nóng)業(yè)生產(chǎn)中,農(nóng)田雜草是影響作物產(chǎn)量的最重要因素之一,雜草會(huì)與作物爭(zhēng)奪陽(yáng)光、肥料、水分和空間,降低作物的產(chǎn)量和品質(zhì),孳生病蟲害,而且會(huì)妨礙農(nóng)事操作。除草劑的應(yīng)用是農(nóng)業(yè)上的一項(xiàng)現(xiàn)代先進(jìn)技術(shù),為防除農(nóng)田雜草起到了極大的推動(dòng)作用。但是,由于作物和部分農(nóng)田雜草的親緣關(guān)系非常近,施用除草劑如何才能做到選擇性地殺死雜草而不傷害農(nóng)作物,已經(jīng)成為除草劑應(yīng)用中所面臨的一個(gè)重要問(wèn)題。針對(duì)這一現(xiàn)狀,發(fā)展抗除草劑作物新品種將是最方便、經(jīng)濟(jì)、有效地控制田間雜草的措施。
目前發(fā)展抗除草劑作物的途徑主要有兩種:一種是通過(guò)轉(zhuǎn)基因手段將抗除草劑基因轉(zhuǎn)入作物中,最成功的案例是美國(guó)孟山都公司開(kāi)發(fā)的一系列轉(zhuǎn)基因抗草甘膦除草劑作物品種農(nóng)達(dá)(Roundup)系列,包括抗農(nóng)達(dá)大豆、玉米、棉花、油菜、向日葵和甜菜。但是利用轉(zhuǎn)基因基因技術(shù)培育的抗除草劑的作物品種需要經(jīng)過(guò)較長(zhǎng)時(shí)間的安全性評(píng)價(jià),而且,從品種選育成功到生產(chǎn)應(yīng)用的周期較長(zhǎng)。第二種是利用遺傳學(xué)非轉(zhuǎn)基因的手段篩選作物抗除草劑突變體。該方法代表了目前國(guó)際抗除草劑作物培育的新方向。首先通過(guò)遺傳誘變的方法構(gòu)建突變體庫(kù),從中篩選到抗除草劑突變體,這樣得到的突變體可以直接作為抗除草劑新品種加以利用,或通過(guò)回交轉(zhuǎn)育的方法雜交到其他栽培品種。其優(yōu)點(diǎn)是非轉(zhuǎn)基因作物,不需要經(jīng)過(guò)安全評(píng)估直接應(yīng)用,產(chǎn)業(yè)化所需的批準(zhǔn)時(shí)間短。因此我們選擇了后者作為獲得抗除草劑作物的方法。
乙酰羥基合成酶(AHAS,EC2.2.1.6)或者叫乙酰乳酸合成酶(ALS)是支鏈氨基酸(纈氨酸、亮氨酸和異亮氨酸)生物合成途徑的第一個(gè)酶,也是幾種重要除草劑的靶位點(diǎn),主要包括:咪唑啉酮類除草劑、磺酰脲類除草劑、三唑并嘧啶類除草劑、嘧啶基氧基苯甲酸酯類除草劑和磺酰基氨基羧基三唑啉酮類除草劑。這五類除草劑與其靶位點(diǎn)乙酰羥基合成酶結(jié)合后,能夠抑制蛋白活性,從而阻止易感植物的生長(zhǎng)發(fā)育。在模式植物擬南芥中的研究表明,AHAS蛋白某些氨基酸突變后能夠使擬南芥獲得除草劑抗性,突變后的蛋白不再與除草劑結(jié)合,同時(shí)又保持原有的生物學(xué)活性。
正是基于以上原理,本發(fā)明人通過(guò)長(zhǎng)期而艱苦的研究實(shí)踐,在冬小麥矮抗58和濟(jì)麥22中構(gòu)建了突變體庫(kù),并從中篩選到一系列抗除草劑突變體,并鑒定出賦予其除草劑抗性的蛋白。本發(fā)明人還開(kāi)發(fā)了這些蛋白及其編碼基因在轉(zhuǎn)基因或者非轉(zhuǎn)基因植物育種中的應(yīng)用,可用于培育具有除草劑抗性的植物,尤其是農(nóng)作物,如抗除草劑小麥等。
技術(shù)實(shí)現(xiàn)要素:
本發(fā)明提供了一種來(lái)源于小麥的IMI核酸序列,所述IMI核酸序列具有咪唑啉酮類除草劑抗性和磺酰脲類除草劑抗性,所述IMI基因的核苷酸序列或氨基酸序列在植物中表達(dá)時(shí)能賦予植物增加的對(duì)咪唑啉酮類除草劑和/或磺酰脲類除草劑的耐受性。在本發(fā)明中,所述IMI核苷酸序列的編號(hào)及其對(duì)應(yīng)的核苷酸和氨基酸序列分別是:抗性基因IMI1-286,其核苷酸序列如SEQ ID NO:1所示,其編碼的氨基酸序列如SEQ ID NO:2所示;抗性基因IMI1-512,其核苷酸序列如SEQ ID NO:3所示,其編碼的氨基酸序列如SEQ ID NO:4所示;抗性基因IMI1-536,其核苷酸序列如SEQ ID NO:5所示,其編碼的氨基酸序列如SEQ ID NO:6所示;抗性基因IMI1-1880,其核苷酸序列如SEQ ID NO:7所示,其編碼的氨基酸序列如SEQ ID NO:8所示;抗性基因IMI2-292,其核苷酸序列如SEQ ID NO:9所示,其編碼的氨基酸序列如SEQ ID NO:10所示;抗性基因IMI2-518,其核苷酸序列如SEQ ID NO:11所示,其編碼的氨基酸序列如SEQ ID NO:12所示;抗性基因IMI2-542,其核苷酸序列如SEQ ID NO:13所示,其編碼的氨基酸序列如SEQ ID NO:14所示;抗性基因IMI2-1886,其核苷酸序列如SEQ ID NO:15所示,其編碼的氨基酸序列如SEQ ID NO:16所示;抗性基因IMI3-295,其核苷酸序列如SEQ ID NO:17所示,其編碼的氨基酸序列如SEQ ID NO:18所示;抗性基因IMI3-521,其核苷酸序列如SEQ ID NO:19所示,其編碼的氨基酸序列如SEQ ID NO:20所示;抗性基因IMI3-545,其核苷酸序列如SEQ ID NO:21所示,其編碼的氨基酸序列如SEQ ID NO:22所示;抗性基因IMI3-1889,其核苷酸序列如SEQ ID NO:23所示,其編碼的氨基酸序列如SEQ ID NO:24所示。
本發(fā)明提供了一種用于增強(qiáng)植物、植物組織、植物細(xì)胞或其他宿主細(xì)胞對(duì)于至少一種除草劑的耐受性或抗性的方法,所述除草劑干擾ALS酶的活性,更具體的所述除草劑包括但不限于磺酰脲類、咪唑啉酮類、嘧啶并三唑類、水楊酸嘧啶類等除草劑(Peter Babczinski,Thomas Zelinski.Mode of action herbicidal ALS-inhibitors on acetolactate synthase from green plant cell cultures,yeast,and Escherichia coli.Pesticide science,1991,31:305-323;鄭培忠,沈健英.乙酰乳酸合成酶抑制劑的種類及其耐藥性研究進(jìn)展.雜草科學(xué),2009,2:4-8)。因此,在本文中,除草劑抗性指的是對(duì)包括磺酰脲類、咪唑啉酮類、嘧啶并三唑類、水楊酸嘧啶類在內(nèi)的除草劑具有耐受性。優(yōu)選在本發(fā)明中,除草劑是咪唑啉酮類除草劑,如在本發(fā)明的具體實(shí)施方式中,除草劑是咪草煙(其化學(xué)名稱為(RS)5-乙基-2-(4-異丙基-4-甲基-5-氧代-3-咪唑啉-2-基)煙酸,CAS號(hào)為81335-77-5)。即優(yōu)選在本發(fā)明中,除草劑抗性是咪唑啉酮類除草劑抗性,更優(yōu)選是咪草煙抗性。
本發(fā)明所述的增強(qiáng)植物、植物組織、植物細(xì)胞或其他宿主細(xì)胞對(duì)至少一種除草劑的耐受性或抗性的方法,可以通過(guò)轉(zhuǎn)化的方法實(shí)施,或是雜交、回交、自交或無(wú)性繁殖等方式實(shí)施,也可以通過(guò)化學(xué)誘變或基因定點(diǎn)突變的方式獲得,使改變后的植株具有本發(fā)明所提供的核苷酸序列SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23。更具體地,所述突變后的植株可以具有一種或一種以上的上述突變后的核苷酸序列。
轉(zhuǎn)化的方法是指通過(guò)農(nóng)桿菌介導(dǎo)的方法,或基因槍法,將本發(fā)明所提供的IMI核苷酸序列SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23轉(zhuǎn)入到植物、植物組織、植物細(xì)胞或其他宿主細(xì)胞中,以增強(qiáng)其對(duì)除草劑的耐受性或抗性?;瘜W(xué)誘變的方法包括用EMS等誘變劑處理所導(dǎo)致的誘變;基因定點(diǎn)突變的方法包括但不限于ZFN定點(diǎn)突變方法、TALEN定點(diǎn)突變方法、和/或CRISPR/Cas9等定點(diǎn)突變方法。
本發(fā)明還提供了一種提高小麥雜交種子純度的方法,包括:通過(guò)轉(zhuǎn)化的方法、化學(xué)誘變、回交轉(zhuǎn)育或定點(diǎn)突變的方法,使得小麥具有本發(fā)明所提供的核苷酸序列SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23,并且同時(shí)具有咪唑啉酮類除草劑和/或磺酰脲類除草劑的抗性或耐受性,將該小麥材料當(dāng)做父本材料,或是將該材料的除草劑抗性回交轉(zhuǎn)育到其他父本材料中,再將父本材料與母本材料進(jìn)行雜交,產(chǎn)生的雜交種子均具有除草劑抗性,而母本材料自己結(jié)實(shí)的種子由于不具備除草劑抗性,將所有結(jié)出的種子在浸種階段用咪唑啉酮類除草劑和/或磺酰脲類除草劑溶液進(jìn)行浸泡,則可以殺死假的雜交種子,或是在秧苗期,對(duì)小苗進(jìn)行除草劑的噴施,殺死非雜交產(chǎn)生的種子秧苗,從而提高雜交種子生產(chǎn)的制種純度,降低雜交種子的種植風(fēng)險(xiǎn)。
本發(fā)明還提供了一種表達(dá)盒(cassette),所述表達(dá)盒包含本發(fā)明所提供的核酸序列SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23。所述表達(dá)盒中的核酸序列由一個(gè)啟動(dòng)子驅(qū)動(dòng)表達(dá),所述啟動(dòng)子包括但不限于CaMV 35S啟動(dòng)子或者稻肌動(dòng)蛋白I基因、玉米醇脫氫酶基因、玉米矮縮I基因或煙草花葉病毒(TMV)Ω元件的啟動(dòng)子。進(jìn)一步的,本發(fā)明所提供的表達(dá)盒還可以包括,在細(xì)菌中復(fù)制所需的復(fù)制起點(diǎn)(例如,來(lái)自pBR322或P15A ori的ORI區(qū)),土壤桿菌T-DNA轉(zhuǎn)移所需要的元件(例如,T-DNA的左邊界和/或右邊界),從而方便從細(xì)菌往植物中的轉(zhuǎn)化過(guò)程。另外,本發(fā)明所述的表達(dá)盒還可以包含增強(qiáng)子、內(nèi)含子、多克隆位點(diǎn)、操縱基因、阻遏物結(jié)合位點(diǎn)、轉(zhuǎn)錄因子結(jié)合位點(diǎn)等,例如來(lái)自TMV、玉米褪綠斑點(diǎn)病毒(MCMV)或苜蓿花葉病毒(AMV)的前導(dǎo)序列等作為增強(qiáng)子,又如可以包括來(lái)自Adh 1、bronze 1、肌動(dòng)蛋白1或肌動(dòng)蛋白2的內(nèi)含子。
本發(fā)明還提供了一種載體,所述載體包含本發(fā)明上述所提供的含有SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23的表達(dá)盒。在本文中,載體是指本領(lǐng)域中常用的細(xì)菌質(zhì)粒、粘粒、噬菌粒、酵母質(zhì)粒、植物細(xì)胞病毒、動(dòng)物病毒及其它各種病毒載體。根據(jù)所應(yīng)用的目的不同,載體可以分為克隆載體、表達(dá)載體和轉(zhuǎn)化載體,指的是所使用的目的分別針對(duì)克隆并驗(yàn)證基因、表達(dá)相應(yīng)基因和將相應(yīng)基因轉(zhuǎn)化。本發(fā)明中可用的載體包括但不限于:在細(xì)菌中表達(dá)用的載體(原核表達(dá)載體)、在酵母中表達(dá)用的載體(如畢赤酵母載體)、在昆蟲細(xì)胞中表達(dá)的桿狀病毒載體、在哺乳動(dòng)物細(xì)胞中表達(dá)用的載體(痘苗病毒載體、逆轉(zhuǎn)錄病毒載體、腺病毒載體、腺伴病毒載體等)、在植物中表達(dá)用的植物病毒載體以及在哺乳動(dòng)物乳腺中表達(dá)用的各種載體。本發(fā)明所述的載體是轉(zhuǎn)化表達(dá)載體,尤其是植物轉(zhuǎn)化載體,特別是適合農(nóng)桿菌轉(zhuǎn)化植物的載體。這些載體很多已經(jīng)商品化了。
本發(fā)明還提供了一種細(xì)胞,所述細(xì)胞包含本發(fā)明所提供的包含SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23序列的表達(dá)盒或載體。細(xì)胞可以是原核細(xì)胞,也可以是真核細(xì)胞,如,細(xì)菌細(xì)胞、酵母細(xì)胞、植物細(xì)胞、昆蟲細(xì)胞、哺乳動(dòng)物細(xì)胞等。在本文中,植物細(xì)胞指的是來(lái)自植物的細(xì)胞(可以包括細(xì)胞群,但優(yōu)選是單個(gè)細(xì)胞),但是這些細(xì)胞不能僅僅借助光合作用而以水、二氧化碳和無(wú)機(jī)鹽等無(wú)機(jī)物合成碳水化合物、蛋白質(zhì)來(lái)發(fā)育成存活的單個(gè)植株。植物細(xì)胞可以是必須使用有機(jī)物(如某些植物激素)才能發(fā)育的植物細(xì)胞或細(xì)胞群,也可以是完全無(wú)發(fā)育成植株能力的植物細(xì)胞或細(xì)胞群,通俗地說(shuō),即死亡的植物細(xì)胞或細(xì)胞群,如干燥粉碎過(guò)的市售小麥面等。
本發(fā)明中所述的植物可以是雙子葉植物,也可以是單子葉植物。示例性的植物包括小麥、水稻、玉米、黑麥、燕麥、大麥、大豆、馬鈴薯、油菜、甜菜、甘蔗、高粱、西紅柿、南瓜、辣椒、生菜、向日葵、咖啡、茶、棉花、苜蓿、煙草、或擬南芥等。在本發(fā)明的具體實(shí)施方式中,優(yōu)選本發(fā)明的植物是小麥,更優(yōu)選是寧春4號(hào)。其中,寧春4號(hào)是我國(guó)在1980年代初期就在寧夏培育出的非轉(zhuǎn)基因小麥品種,已經(jīng)可以在我國(guó)推廣使用了30年,是我國(guó)解放以來(lái)累計(jì)種植面積最大的小麥品種,其品種審定編號(hào)為寧審種8101。本發(fā)明優(yōu)選的植物,可以繼承了寧春4號(hào)豐產(chǎn)性突出、適應(yīng)性廣泛、品質(zhì)優(yōu)良等特性,并具有除草劑抗性,尤其具有咪唑啉酮類除草劑抗性,特別是咪草煙抗性。
本發(fā)明還提供了本發(fā)明上述方法或核苷酸序列、氨基酸序列在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用。在獲得具有除草劑抗性的植物過(guò)程中,可以應(yīng)用到本發(fā)明前述各方面所述的方法中。本發(fā)明所提供的還包括,本發(fā)明所提供的蛋白質(zhì)SEQ ID NO:2、SEQ ID NO:4、SEQ ID NO:6、SEQ ID NO:8、SEQ ID NO:10、SEQ ID NO:12、SEQ ID NO:14、SEQ ID NO:16、SEQ ID NO:18、SEQ ID NO:20、SEQ ID NO:22和/或SEQ ID NO:24在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用,本發(fā)明所提供的核酸SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用,本發(fā)明所提供的表達(dá)盒在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用,本發(fā)明所提供的載體在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用,本發(fā)明所提供的細(xì)胞在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用,本發(fā)明所提供的植物在獲得具有除草劑抗性的植物過(guò)程中的應(yīng)用。優(yōu)選在本發(fā)明中,除草劑是咪唑啉酮類除草劑和磺酰脲類除草劑,優(yōu)選是咪草煙。
在本文中,獲得包括制備、培育、生產(chǎn)或以其他方式產(chǎn)生得到,包括通過(guò)轉(zhuǎn)基因育種方式獲得,如將本發(fā)明所提供的核酸SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23轉(zhuǎn)化入植物后使之表達(dá)本發(fā)明的蛋白質(zhì);也包括通過(guò)非轉(zhuǎn)基因育種方式獲得,如通過(guò)雜交、回交、自交或無(wú)性繁殖本發(fā)明所獲得的植物并分選出仍舊包含本發(fā)明所提供的核酸并表達(dá)本發(fā)明所提供的蛋白質(zhì)的植物。因此,本發(fā)明還提供了一種應(yīng)用,包括:在獲得具有除草劑抗性的轉(zhuǎn)基因植物過(guò)程中的應(yīng)用和在獲得具有除草劑抗性的非轉(zhuǎn)基因植物過(guò)程中的應(yīng)用。
本發(fā)明還提供了獲得具有除草劑抗性的植物的方法,其包括如下步驟:
(1)使植物包含本發(fā)明所提供的核酸SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23;和/或,
(2)使植物表達(dá)本發(fā)明所提供的蛋白質(zhì)SEQ ID NO:2、SEQ ID NO:4、SEQ ID NO:6、SEQ ID NO:8、SEQ ID NO:10、SEQ ID NO:12、SEQ ID NO:14、SEQ ID NO:16、SEQ ID NO:18、SEQ ID NO:20、SEQ ID NO:22和/或SEQ ID NO:24。
此外,本發(fā)明還提供了一種鑒定本植物中是否含有本發(fā)明核苷酸序列的方法,其包括如下步驟:
(1)測(cè)定所述植物是否包含本發(fā)明所提供的核酸SEQ ID NO:1、SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21和/或SEQ ID NO:23;和/或,
(2)測(cè)定所述植物是否表達(dá)本發(fā)明所提供的蛋白質(zhì)SEQ ID NO:2、SEQ ID NO:4、SEQ ID NO:6、SEQ ID NO:8、SEQ ID NO:10、SEQ ID NO:12、SEQ ID NO:14、SEQ ID NO:16、SEQ ID NO:18、SEQ ID NO:20、SEQ ID NO:22和/或SEQ ID NO:24。
測(cè)定的步驟可以通過(guò)常規(guī)的核酸檢測(cè)和/或蛋白質(zhì)檢測(cè)方法來(lái)進(jìn)行,只需要能夠檢測(cè)出蛋白質(zhì)或其編碼核酸帶有相應(yīng)核酸突變的方法都可以,示例性的方法包括蛋白質(zhì)測(cè)序、核酸測(cè)序、聚合酶鏈?zhǔn)椒磻?yīng)(PCR)檢測(cè)、探針雜交檢測(cè)等。
本發(fā)明取得的有益效果在于獲得可替代現(xiàn)有技術(shù)的使植物具有除草劑抗性的蛋白質(zhì)、核酸、表達(dá)盒、載體、細(xì)胞、植物、獲得具有除草劑抗性的植物的應(yīng)用和方法、以及鑒定本發(fā)明的植物的方法,并且可以通過(guò)轉(zhuǎn)基因或非轉(zhuǎn)基因方法獲得具有除草劑抗性的植物品種,尤其是能繼承寧春4號(hào)優(yōu)良特性的小麥品種。

具體實(shí)施方式
下述實(shí)施例中所用方法如無(wú)特別說(shuō)明,均為常用分子生物學(xué)、組織培養(yǎng)技術(shù)和農(nóng)學(xué)手冊(cè)所記載的方法。例如,具體步驟可參見(jiàn):《Molecular Cloning:A Laboratory Manual(3rd edition)》(Sambrook,J.,Russell,David W.,2001,Cold Spring Harbor),《Plant Propagation by Tissue Culture》(Edwin F.George,Michael A.Hall,Geert-Jan De Klerk,2008,Springer)。
實(shí)施例1.小麥突變體庫(kù)的構(gòu)建及抗除草劑突變體的篩選
取市售的小麥品種——春小麥寧春四號(hào)種子400kg、冬小麥矮抗58和濟(jì)麥22種子各20kg,用水浸沒(méi)種子4小時(shí),將種子撈出,控干水分。然后,用0.5%(w/w)甲磺酸乙酯(EMS)浸泡種子12小時(shí),保證液面浸沒(méi)種子。12小時(shí)后棄去EMS溶液,換自來(lái)水浸泡種子,棄自來(lái)水,浸泡共重復(fù)5次,每次浸泡5分鐘,然后用自來(lái)水沖洗種子2小時(shí),沖洗過(guò)程中翻動(dòng)種子,從而將EMS洗凈,稍微風(fēng)干后播種,待植株成熟后收割,其中春小麥寧春四號(hào)收獲M2代(突變體庫(kù)2代)種子3500公斤,矮抗58和濟(jì)麥22兩個(gè)品種各收獲120公斤。
將M2代種子用濃度為500毫克/升的咪唑啉酮類除草劑咪草煙((RS)5-乙基-2-(4-異丙基-4-甲基-5-氧代-3-咪唑啉-2-基)煙酸,CAS號(hào)81335-77-5)在23度環(huán)境下浸泡18小時(shí),然后將除草劑倒掉,將種子均勻地鋪撒在沙床上萌發(fā)。3周后,發(fā)現(xiàn)除草劑浸泡處理的種子中有綠色秧苗持續(xù)生長(zhǎng)(圖1),其中矮抗58品種獲得119株疑似陽(yáng)性植株,濟(jì)麥22獲得117株疑似陽(yáng)性植株。
實(shí)施例2.抗除草劑陽(yáng)性植株的分子水平鑒定
為了證明本發(fā)明獲得的除草劑抗性突變體與植株體內(nèi)的乙酰羥基合成酶突變的關(guān)系,本實(shí)施例對(duì)獲得的抗除草劑陽(yáng)性植株進(jìn)行了分子水平的鑒定。
小麥基因組中的三個(gè)乙酰羥基合成酶基因IMI1、IMI2和IMI3分別位于D基因組、B基因組和A基因組中。取篩選獲得的疑似抗除草劑陽(yáng)性植株葉片,連同野生型植株葉片,分別單株提取基因組DNA,并以基因組DNA為模板將小麥基因組中編碼的3個(gè)乙酰羥基合成酶IMI1、IMI2和IMI3通過(guò)聚合酶鏈?zhǔn)椒磻?yīng)擴(kuò)增其全長(zhǎng)基因組DNA序列,擴(kuò)增引物分別是:
TaImi1-F:TAAAGAAGCTAGCCCAATCTGAAC(SEQ ID NO:25)和TaImi1-R:AGAACATAGACTACAAAACTACAGG(SEQ ID NO:26)用于擴(kuò)增IMI1基因;
TaImi2-F:CACAACCGCGAGTGGAAATACC(SEQ ID NO:27)和TaImi2-R:GAGAACAGAGACTACAAAACTACACG(SEQ ID NO:28)用于擴(kuò)增IMI2基因;
TaImi3-F:GTTTGAGCATCAGCCTTTCTGCAC(SEQ ID NO:29)和
TaImi3-R:TCCATCGGTCATTATGTCATAACCG(SEQ ID NO:30)用于擴(kuò)增IMI3基因。
PCR結(jié)束后,通過(guò)電泳膠回收,將獲得的回收液委托北京三博遠(yuǎn)志生物技術(shù)有限公司進(jìn)行測(cè)序,并對(duì)測(cè)序結(jié)果進(jìn)行分析。分析結(jié)果顯示,小麥由于是異源六倍體(AABBDD),其三個(gè)拷貝的乙酰羥基合成酶基因的序列同源性非常高,其中IMI1與IMI2之間的核苷酸和蛋白質(zhì)一致性分別為97%和99%,IMI1與IMI3之間的核苷酸和蛋白質(zhì)一致性分別為98%和98%,IMI2與IMI3之間的核苷酸和蛋白質(zhì)一致性分別為97%和98%。
分析結(jié)果發(fā)現(xiàn),在鑒定的236株陽(yáng)性植株中,有229株發(fā)現(xiàn)了3個(gè)乙酰羥基合成酶中某一位點(diǎn)發(fā)生了突變,在3個(gè)基因上各有4個(gè)突變位點(diǎn)(見(jiàn)圖2),分別是IMI1第286位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第96位丙氨酸變?yōu)樘K氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:1所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:2所示;IMI1第512位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第171位脯氨酸變?yōu)榱涟彼幔撏蛔冎仓甑腁LS基因的核苷酸序列如SEQ ID NO:3所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:4所示;IMI1第536位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第179位丙氨酸變?yōu)槔i氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:5所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:6所示;IMI1第1880位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第627位絲氨酸變?yōu)樘於0罚撏蛔冎仓甑腁LS基因的核苷酸序列如SEQ ID NO:7所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:8所示。IMI2第292位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第98位丙氨酸變?yōu)樘K氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:9所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:10所示;IMI2第518位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第173位脯氨酸變?yōu)榱涟彼?,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:11所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:12所示;IMI2第542位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第181位丙氨酸變?yōu)槔i氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:13所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:14所示;IMI2第1886位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第629位絲氨酸變?yōu)樘於0罚撏蛔冎仓甑腁LS基因的核苷酸序列如SEQ ID NO:15所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:16所示。IMI3第295位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第99位丙氨酸變?yōu)樘K氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:17所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:18所示;IMI3第521位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第174位脯氨酸變?yōu)榱涟彼?,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:19所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:20所示;IMI3第545位由C到T的突變,導(dǎo)致相應(yīng)編碼氨基酸由第182位丙氨酸變?yōu)槔i氨酸,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:21所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:22所示;IMI3第1889位由G到A的突變,導(dǎo)致相應(yīng)編碼氨基酸由第630位絲氨酸變?yōu)樘於0?,該突變植株的ALS基因的核苷酸序列如SEQ ID NO:23所示,其編碼的ALS蛋白的氨基酸序列如SEQ ID NO:24所示。
實(shí)施例3.抗除草劑陽(yáng)性植株后代田間咪唑啉酮類除草劑的耐受表型
將鑒定為純合突變體的株系后代單株收種,在田間種約一平方米的小區(qū),同時(shí)旁邊種植野生型作為對(duì)照。待小麥長(zhǎng)至三葉期時(shí),噴施以濃度為500毫克/升的咪草煙,濃度是商用推薦劑量的2倍。噴施一個(gè)月后調(diào)查秧苗枯死情況,我們發(fā)現(xiàn)野生型對(duì)照在噴施咪草煙之后幾乎停止生長(zhǎng),新長(zhǎng)出來(lái)的葉片完全枯黃,進(jìn)而逐漸枯死,而我們篩選到的不同位點(diǎn)的抗除草劑陽(yáng)性苗則全部呈現(xiàn)對(duì)咪草煙完全耐受的表型,其正常的生長(zhǎng)發(fā)育幾乎沒(méi)有受到影響(見(jiàn)圖3和圖4),最終正常拔節(jié)抽穗。這表明我們篩選到的抗除草劑突變體確實(shí)能夠穩(wěn)定遺傳地獲得對(duì)咪唑啉酮類除草劑的耐受。
實(shí)施例4.抗性材料的兩兩雜交
為了檢測(cè)是否具有雙重或雙重以上突變位點(diǎn)的抗性植株會(huì)比具有單位點(diǎn)抗性植株的除草劑耐受能力更強(qiáng),我們將實(shí)施例1和2中獲得的具有不同突變位點(diǎn)的抗性植株進(jìn)行了兩兩雜交。
選擇IMI1-G1880A突變位點(diǎn)分子鑒定為純合的株系G38(序列對(duì)應(yīng)SEQ ID NO:7)、IMI2-G1886A突變位點(diǎn)分子鑒定為純合的株系E88(序列對(duì)應(yīng)SEQ ID NO:15)以及IMI3-G1889A突變位點(diǎn)分子鑒定為純合的株系E14(序列對(duì)應(yīng)SEQ ID NO:23),將這三個(gè)株系在溫室種植,開(kāi)花期將G38與E88、G38與E14、E88與E14進(jìn)行雜交。具體方法是:先對(duì)母本穗提前進(jìn)行人工去雄并套袋,父本植株開(kāi)花的時(shí)候,將父本穎殼剪掉,光下放置約10分鐘,待花藥從穎殼伸長(zhǎng)出來(lái)后,將父本花粉抖到母本穗上,重復(fù)兩次,待種子成熟后收種,即為F1代雜交種。收獲的F1代雜交種繁殖一代,收獲F2代群體,在G38與E88雜交F2代群體中可以鑒定到同時(shí)帶有純合的IMI1-G1880A和IMI2-G1886A突變的植株,在G38與E14雜交F2代群體中可以鑒定到同時(shí)帶有純合的IMI1-G1880A和IMI3-G1889A突變的植株,在E888與E14雜交F2代群體中可以鑒定到同時(shí)帶有純合的IMI2-G1886A和IMI3-G1889A突變的植株。用除草劑對(duì)這些帶有雙位點(diǎn)突變的純合植株進(jìn)行噴施實(shí)驗(yàn),結(jié)果表明,同時(shí)突變兩個(gè)位點(diǎn)的植株相對(duì)于只有一個(gè)突變位點(diǎn)的植株,對(duì)除草劑的耐受性得到了顯著的提高。
