背景技術(shù):
:水稻作為我國(guó)第一大糧食作物,隨著人民生活水平的提高及消費(fèi)觀念的改變,人們對(duì)稻米品質(zhì)有了更高的要求。稻米堊白是評(píng)價(jià)稻米品質(zhì)的重要指標(biāo):(1)稻米堊白是一個(gè)非常重要的外觀品質(zhì)性狀(Tan等,2000,Theor.Appl.Genet.105:248-257)。在現(xiàn)在的稻米市場(chǎng)上,消費(fèi)者對(duì)稻米的選擇不僅會(huì)考慮到口味的舒適,還很關(guān)注外觀的優(yōu)劣,因此稻米的外觀品質(zhì)直接決定了稻米的商業(yè)價(jià)值。(2)稻米堊白會(huì)直接影響稻米外觀品質(zhì)、碾磨加工品質(zhì)和蒸煮食味品質(zhì),是稻米最重要的品質(zhì)性狀之一(Fitzgeraldetal,2009,TrendsPlantSci.14:133-139)。因此闡明稻米堊白的遺傳基礎(chǔ)和分子機(jī)理有利于稻米品質(zhì)的遺傳改良。堊白是指稻米胚乳中白色不透明的部分,是因?yàn)榕呷橹械牡矸垠w與蛋白質(zhì)體排列疏松,相互間存在著空氣,陽(yáng)光照射胚乳導(dǎo)致光線折射所形成的一種光學(xué)特性(Yamakawaetal,2007,PlantPhysiol.144:258–277;Zhang,2007,Proc.Natl.Acad.Sci.104:16402-16409)。按照?qǐng)装自谧蚜I习l(fā)生部位的不同,可分為腹白、心白和背白,通常以堊白粒率、堊白面積和堊白度來(lái)表示(程方民等,2000,中國(guó)水稻科學(xué).14:83-87;Yamakawaetal,2007,PlantPhysiol.144:258–277)。有堊白的稻米存在部分不透明現(xiàn)象,易影響其外觀品質(zhì)與流通;在加工過(guò)程中由于堊白部分與透明部分的硬度不同,易導(dǎo)致米粒斷裂,從而降低整精米率,影響碾磨品質(zhì);蒸煮后的飯粒易產(chǎn)生裂紋或蓬松中空,影響其蒸煮食味品質(zhì)(Yamakawaetal,2007,PlantPhysiol.144:258–277;Zhang,2007,Proc.Natl.Acad.Sci.104:16402-16409)。目前,堊白與整精米率是國(guó)際市場(chǎng)中決定稻米市場(chǎng)價(jià)格的兩個(gè)最重要因素,并且堊白對(duì)整精米率的高低有間接的影響。所以,為了提高我國(guó)稻米的競(jìng)爭(zhēng)力、滿足市場(chǎng)和消費(fèi)者的需求,稻米品質(zhì)的改良成為我國(guó)稻米產(chǎn)業(yè)發(fā)展的重點(diǎn)。盡管到目前為止已經(jīng)克隆了一些與堊白相關(guān)的基因,但這些基因還遠(yuǎn)遠(yuǎn)不能滿足基因工程改良稻米品質(zhì)的需求,因此本發(fā)明分離到的控制稻米胚乳堊白的基因?qū)Φ久灼焚|(zhì)的改良有重要意義。技術(shù)實(shí)現(xiàn)要素:針對(duì)現(xiàn)有技術(shù)存在的問(wèn)題,本發(fā)明從水稻中分離并克隆一個(gè)控制稻米堊白基因的完整片段及其CDS片段,旨在利用此基因提高改良稻米品質(zhì)的能力。申請(qǐng)人將克隆的該基因命名為OsPK2。其中,本發(fā)明所述片段如序列表SEQIDNO:1-6所示,或者基本上相當(dāng)于SEQIDNO:1-6所示的高度同源序列。具體地,本發(fā)明分離克隆的基因及上述基因的氨基酸序列的信息如下所示:1)該基因的核苷酸序列如序列表SEQIDNO.1所示,其CDS序列如序列表SEQIDNO.2所示;2)其編碼蛋白的氨基酸序列如序列表SEQIDNO.3所示;3)其突變體基因的核苷酸序列如序列表SEQIDNO.4所示,CDS序列如序列表SEQIDNO.5所示;4)其突變體基因編碼蛋白的氨基酸序列如序列表SEQIDNO.6所示.所述的一種轉(zhuǎn)基因細(xì)胞系,其特征在于含有如SEQIDNO.1所示的核苷酸序列。所述的一種轉(zhuǎn)基因重組菌,其特征在于含有如SEQIDNO.1所示的核苷酸序列。所述的編碼丙酮酸激酶蛋白質(zhì)的基因在培育水稻胚乳堊白變化的轉(zhuǎn)基因植物中的應(yīng)用。本發(fā)明的優(yōu)點(diǎn)在于:本發(fā)明克隆了一個(gè)控制稻米堊白的基因,為水稻等禾谷類作物的優(yōu)質(zhì)育種提供了新的基因資源,也為其他作物中克隆相關(guān)基因提供了技術(shù)支持。附圖說(shuō)明圖1.野生型和突變體糙米表型圖,掃描電鏡圖以及粒形和粒重統(tǒng)計(jì)。圖2.本發(fā)明的OsPK2基因的圖位克隆。圖A為OsPK2精細(xì)定位圖;圖B是OsPK2的基因結(jié)構(gòu)。圖3.野生型與突變體的理化性質(zhì)測(cè)定。圖4.OsPK2基因在水稻全生育期時(shí)空表達(dá)譜分析。圖5.本發(fā)明用到的雙元載體pCAMBIA1300的載體圖。圖6.本發(fā)明的T1代互補(bǔ)轉(zhuǎn)基因陽(yáng)性單株的表型圖以及粒形和粒重統(tǒng)計(jì)。具體實(shí)施方式為了理解本發(fā)明,下面以實(shí)施例進(jìn)一步說(shuō)明本發(fā)明,但不限制本發(fā)明。實(shí)施例1:OsPK2基因的圖位克隆1)水稻材料水稻(OryzaSativaL)突變體ospk2(pyruvatekinase2),原始野生型材料為粳稻品種中花11(ZH11)。2)理化性質(zhì)測(cè)定(參考國(guó)標(biāo))直鏈淀粉含量的測(cè)定稱量出0.0500g的米粉置于50ml的容量瓶中,輕微震蕩容量瓶將容量瓶管壁附著的米粉震蕩至瓶底。先加入500μl的無(wú)水乙醇,輕微混勻,再沿容量瓶瓶頸加入4.5ml的1mol/L的NaOH溶液,室溫下靜置23h。然后用去離子水定容至50ml,搖晃均勻。吸取0.5ml樣品和5ml水至試管中,在試管中加入100μl的1mol/L的醋酸溶液,200μl的KI-I2和4.2ml的H2O,振蕩器劇烈震蕩混勻,靜置20min。用直鏈淀粉含量分別為1.5%、10.4%、16.2%和26.5%的標(biāo)準(zhǔn)樣品繪制標(biāo)準(zhǔn)曲線。用空白溶液于分光光度計(jì)波長(zhǎng)620nm處調(diào)節(jié)零點(diǎn)并測(cè)出有色樣品液的吸光度值。然后用標(biāo)準(zhǔn)曲線計(jì)算樣品中直鏈淀粉含量。蛋白含量測(cè)定稱取樣品約2.00g(±0.001g),移入干燥的100ml凱氏燒瓶中,加入0.2g硫酸銅和6g硫酸鉀,稍搖勻后瓶口使用小漏斗,加入20mL濃硫酸,傾斜放置石棉網(wǎng)上,在通風(fēng)櫥中加熱消化,開(kāi)始時(shí)用低溫加熱,待內(nèi)容物全部炭化,泡沫停止后,再升高溫度保持微沸,消化至液體呈藍(lán)綠色澄清透明后,繼續(xù)加熱0.5h,取下放冷,小心加20mL水,放冷后,無(wú)損地轉(zhuǎn)移到100ml容量瓶中,加水定容至刻度,混勻備用,即為消化液。試劑空白實(shí)驗(yàn):取與樣品消化相同的硫酸銅、硫酸鉀、濃硫酸,按以上同樣方法進(jìn)行消化,冷卻,加水定容至100ml,得試劑空白消化液。脂肪含量測(cè)定將索氏提取器用蒸餾水清洗后烘干。脂肪燒瓶在120°C的烘箱內(nèi)干燥至恒重(前后兩次稱量差不超過(guò)2mg)。記錄重量。準(zhǔn)確稱取米粉樣品2g(精確至0.01mg),裝入濾紙筒內(nèi)。記錄重量。樣品測(cè)定(1)將濾紙筒放入索氏提取器的抽提筒內(nèi),連接已干燥至恒重的脂肪燒瓶,由抽提器冷凝管上端加入乙醚或石油醚至瓶?jī)?nèi)容積的2/3處,通入冷凝水,將瓶底浸沒(méi)在水浴中加熱,用一小團(tuán)脫脂棉輕輕塞入冷凝管上口。(2)抽提溫度的控制:水浴溫度應(yīng)控制在使提取液在每6-8min回流一次為宜。(3)抽提時(shí)間的控制:抽提時(shí)間為6小時(shí)。(4)提取完畢。取下脂肪燒瓶,回收乙醚或石油醚。待燒瓶?jī)?nèi)乙醚僅剩下1-2ml時(shí),在水浴上趕盡殘留的溶劑,于95-105°C下干燥2h后,置于干燥器中冷卻至室溫,稱量。繼續(xù)干燥30min后冷卻稱量,反復(fù)干燥至恒重(前后兩次稱量差不超過(guò)2mg)。記錄重量。3)掃描電鏡(SEM)觀察野生型ZH11和突變體ospk2完整的干燥種子,去殼成糙米后用小刀輕輕在糙米上刻一圈,用手掰開(kāi),利用黑色導(dǎo)電膠將斷裂部位粘著于銅樣臺(tái)上,用真空鍍膜儀對(duì)自然橫斷面鍍金,采用20kV的加速電壓,于掃描電鏡下對(duì)米粒橫截面的不同部位進(jìn)行觀察和拍照(康海岐等,2007)。發(fā)現(xiàn)突變體種子胚乳中央不透明部位淀粉粒呈疏松圓形排列,而邊緣透明部位的淀粉粒表現(xiàn)為排列緊密的多邊體結(jié)構(gòu);野生型糙米不論是中央還是邊緣部位淀粉粒均表現(xiàn)為致密的多邊體結(jié)構(gòu)(圖1)。4)遺傳分析和定位群體本研究對(duì)突變體ospk2的表型進(jìn)行了遺傳分析。突變體ospk2分別與野生型ZH11以及秈稻NanJing11(NJ11)雜交得到的F1種子胚乳均表現(xiàn)為透明。突變體ospk2與秈稻NJ11雜交所得的F2群體中,透明胚乳表型與堊白胚乳表型出現(xiàn)分離,透明表型和堊白表型種子的分離比符合3:1(見(jiàn)表1)。因此,遺傳分析結(jié)果表明突變體ospk2的表型符合單隱性核基因的遺傳分離規(guī)律。選取突變體和NJ11進(jìn)行雜交,F(xiàn)1代自交,單株收種F2群體,從有分離的群體中選出978個(gè)隱性個(gè)體作為定位群體。每株取1克左右的葉片,用來(lái)提取總DNA進(jìn)行基因定位。表1F2群體中堊白胚乳和透明胚乳種子的分離情況5)OsPK2基因的初步定位和精細(xì)定位本實(shí)驗(yàn)采用SDS法(DellaportaSL,WoodJ,HicksJB,1983,AplantDNAminipreparation:versionII.PlantMol.Biol.Rep.1:19-21)提取水稻樣品基因組DNA。(1)磨樣:將葉片剪碎后裝入2ml的離心管中,大概裝入1/4左右離心管的高度,然后把一粒鋼珠放入管內(nèi),蓋好管蓋,將離心管放入液氮中冷凍約30s~1min后迅速固定在打樣機(jī)上以23~26轉(zhuǎn)/S的速度振動(dòng)20s左右至樣品成粉末狀即可。(2)加入600μlSDS提取液至離心管中,搖勻,65℃水浴30min,其間震蕩混勻2次。(3)打開(kāi)離心管,加入150μlKAC(1/4體積),搖勻后放入冰中大約20min。(4)加入750μl氯仿:異戊醇混合液(24:1),搖床上充分搖勻(120rpm、30min)后于12000rpm離心5min。(5)吸取400μl上清液至事先準(zhǔn)備好的1.5ml離心管中,加入事先在-20℃冰箱中預(yù)冷的無(wú)水乙醇1mL,搖勻后,-20℃冰箱靜置20min后離心(12000rpm、6min)。(7)將上層無(wú)水乙醇倒入廢水缸中,75%乙醇清洗沉淀,風(fēng)干后加入400μlddH2O,-20℃保存?zhèn)溆?。?μlDNA作
