      [0002]流行病學(xué)調(diào)查和實(shí)驗(yàn)室研究證明自然界和人為制造的空氣食品污染,神經(jīng)毒,各種殺蟲劑,重金屬,物理,化學(xué),生物,藥物,社會(huì),精神,情緒,心理等惡性刺激,以及酗酒抽煙,營(yíng)養(yǎng)不良,營(yíng)養(yǎng)不平衡,不良生活習(xí)慣,衰老,等等,都是癌癥和多種其他疾病的危險(xiǎn)因素。這些危險(xiǎn)因素同時(shí)也危害廣泛大量分布于人和動(dòng)物體內(nèi)血,組織和細(xì)胞中的膽堿脂酶(ChE)包括乙酰膽堿脂酶(AChE)和假性膽堿脂酶(BChE)蛋白酶結(jié)構(gòu),類型,和活性及其基因功能結(jié)構(gòu)的完整性。
      [0003]AChE的作用是極其快速地水解神經(jīng)源性和非神經(jīng)源性乙酰膽堿(Ach),研究顯示,每分子AChE每秒鐘可以降解約25,000分子的Ach接近于彌漫底物的程度;由于人體血和受累組織細(xì)胞中的AChE蛋白及其基因逐漸被危險(xiǎn)因素抑制破壞,組織細(xì)胞為了存活就釋放過(guò)量乙酰膽堿(Ach),功能和結(jié)構(gòu)受損的AChE無(wú)法快速充分水解Ach,導(dǎo)致AChE和Ach平衡失調(diào),Ach在受累組織細(xì)胞內(nèi)外過(guò)剩。通常,業(yè)內(nèi)認(rèn)為Ach只是一種神經(jīng)介質(zhì),主要存在于神經(jīng)組織和神經(jīng)肌肉接頭處,近年來(lái)非神經(jīng)性的ACh已經(jīng)被證實(shí)存在,幾乎分布于每個(gè)細(xì)胞,屬于體內(nèi)自分泌旁分泌激素,具有強(qiáng)大的促生長(zhǎng)和求生存作用。研究證實(shí)煙草中的尼古丁和其他有害成分具有Ach作用并可以強(qiáng)烈刺激Ach分泌。過(guò)剩的Ach能異常激活廣泛分布在細(xì)胞膜上的Ach受體,鈣,鉀通道,cAMP, cGMP等一系列第二信號(hào)系統(tǒng)及其因子,達(dá)到刺激癌細(xì)胞生長(zhǎng)和轉(zhuǎn)移。近年來(lái)Ach濃度和/或Ach合成酶,輸送酶及其mRNA表達(dá)水平在人和/或?qū)嶒?yàn)室的肺,肝,結(jié)腸腫瘤細(xì)胞,組織,腫瘤動(dòng)物模型中證實(shí)是顯著升高的,而hAchE/hBChE基因和/或mRNA表達(dá)改變,酶活性水平明顯下降,酶蛋白型,酶糖化型,等等多種改變?cè)趷盒詫?shí)體瘤包括長(zhǎng)在腦(腦膜瘤,腦膠質(zhì)瘤),呼吸道(多種肺癌),消化道(食道,胃,腸,肝,胰腺),生殖泌尿系統(tǒng)(腎,膀胱,宮頸,卵巢,乳房,前列腺),皮膚,軟組織,和血液系統(tǒng)腫瘤(白血病,淋巴細(xì)胞腫瘤等等)均有報(bào)道。平衡失調(diào)的AChE和Ach系統(tǒng)可引起受累組織細(xì)胞的代謝,信號(hào)傳導(dǎo),細(xì)胞周期,凋亡,染色體和其基因等的正常調(diào)控失調(diào),導(dǎo)致正常細(xì)胞慢慢轉(zhuǎn)化成癌細(xì)胞和癌組織并生長(zhǎng)轉(zhuǎn)移最后奪取生命。本申請(qǐng)的發(fā)明人于2008年在國(guó)際首次以醫(yī)學(xué)假說(shuō)形式提出了有關(guān)AChE和Ach上游平衡失調(diào)引起疾病以及用補(bǔ)充AChE/BChE來(lái)預(yù)防和治療疾病的認(rèn)知。目前主流科學(xué)人員仍然側(cè)重對(duì)受體及其激動(dòng)劑和拮抗劑對(duì)疾病治療的研究。
